Polymerase Chain Reaction (PCR)

Test Overview

Duration: Variable

Polymerase Chain Reaction (PCR) is a molecular testing method used to detect specific DNA sequences. At the Iowa State University Seed Laboratory, PCR may be used for both biotech trait testing and the detection or identification of certain seedborne pathogens.

PCR is highly sensitive and can detect very small amounts of target DNA. Depending on the test and testing objective, results may be qualitative (detected/not detected) or quantitative.

Biotech Trait Testing

PCR can be used to detect DNA associated with biotech traits. It is particularly useful when testing seed for the presence or absence of genetically modified material or when a specific trait or genetic sequence must be detected.

The appropriate test and sample size depend on the crop, target, and level of detection required.

Seed Health Testing

PCR may also be used to detect or confirm certain seedborne pathogens. The method targets DNA sequences specific to the organism being tested and may be used alone or as a confirmation method following another seed health test.

Test Details

Seed or plant material is processed and DNA is extracted from the sample. PCR is then used to amplify and detect DNA sequences specific to the target trait or pathogen.

Different PCR methods may be used depending on the testing objective. Results are interpreted using appropriate controls and are reported according to the specific test performed.

Sample Requirements

Sample requirements vary considerably depending on the crop, target, testing objective, and required level of detection.

Depending on the test requested, approximately 1,000–30,000 seeds may be required.

Contact Customer Care if you are unsure of the amount of seed required for your specific test. Additional seed may be required when multiple tests are requested on the same sample.
